Chemopreventive potential of canola and mustard on colon tumorigenesis in Fisher 344 male rats

Canola/mustard sprouts and seeds (Brassica napus and Brassica campestris) are rich sources of omega-3 (n-3) PUFA and other phytochemicals such as isothiocyanates and indoles. The chemopreventive potential of canola/mustard seeds (S) and canola sprouts (SP) at 5% level was tested against azoxymethane (AOM)-induced tumor formation in Fisher 344 male rats. Following a 1wk period of acclimatization, Fisher 344 male rats were randomly assigned to 7 groups (n=12). One group of was fed an AIN93G (control-C diet). Three groups were fed three varieties of S: C+10% Jetton (JS), C+10% Ida Gold (IS) and C+10% Pacific Gold (PS) and SP: C+ 10% Jetton (JSP), C+10% Ida Gold (ISP), and C+10% Pacific Gold (PSP), respectively. At 7 and 8 wk of age all rats received 2 successive s/c injections of AOM in saline at 16mg/kg body weight. Rats were killed by CO2 asphyxiation at 45 wk of age. Tumor incidence (%) in rats fed C was 91.67. Rats fed JS, IS, PS, JSP, ISP, PSP had a tumor incidence was 50, 50, 64, 42, 50, and 64, respectively. The number of tumors in the rats fed C, JSP, ISP, PSP, JS, IS, and PS were 25, 6, 9, 10, 9, 6, and 10. Tumors per tumor-bearing ratios (TBR) for groups fed C , JS, IS, PS, JSP, ISP, PSP, were 2.2, 1.5, 1.0, 1.4, 1.2, 1.5, and 1.6, respectively. Glutathione S-transferase activity (┬Ámol/mg) ranged between 23 and 29 for the control and canola/mustard groups, there were no significant differences among control and treatment groups. The results from this study show that canola and mustard (seeds and sprouts) reduced the formation of AOM induced colon tumors and may possibly have applications in functional food-product development and have implications as a chemopreventive agent.
